Tempest Therapeutics

Tempest Therapeutics is a clinical-stage biotechnology company developing targeted and immune-mediated small-molecule and in vivo CAR-T therapeutics for cancer and autoimmune disease. Its lead candidate, amezalpat (TPST-1120), is an oral selective antagonist of peroxisome proliferator-activated receptor alpha (PPARα) that has completed a Phase 2 trial in first-line unresectable or metastatic hepatocellular carcinoma and is advancing into a pivotal Phase 3 study in combination with atezolizumab and bevacizumab, with Roche supplying atezolizumab and Tempest sponsoring the trial. The pipeline also includes TPST-1495, a dual EP2/EP4 prostaglandin E2 receptor antagonist, and TPST-4003, a CD7-targeted mRNA/LNP in vivo CAR-T candidate. In September 2026 the company signed an exclusive option agreement with Hebei Senlang Biotechnology covering a clinical-stage CD7-targeted lentiviral in vivo CAR-T platform and an associated multiple myeloma Phase 1 asset, following a July 2026 investigator-initiated-trial collaboration between the two companies.
